Behbehani, Abbas M. (Baylor University College of Medicine, Houston, Tex.), and L. H. Lee. Growth, plaque production, and cationic stabilization of rhinovirus type 1 (echovirus 28). J. Bacteriol. 88:1608-1611. 1964.-A variety of conditions for the growth of echovirus 28 were investigated. The virus grew more readily and to the highest titer when secondary MK cells maintained in low bicarbonate and rolled at 33 C were employed. Viral plaques were produced in human diploid (WI-38) cell cultures. The virus was protected from thermal inactivation (50 C for 1 hr) by 1 m MgCl(2). For storage of the virus at refrigerator and room temperature, the use of 1 m MgCl(2) is also of advantage.

Antiviral activity and mechanism of action of 2-(3,4-dichlorophenoxy)-5-nitrobenzonitrile (MDL-860).
A nitrobenzene derivative, MDL-860, was found to inhibit plaque formation, cytopathic effect, or both in 11 of 12 picornaviruses at concentrations which did not affect cell growth. The compound did not directly inactivate the virus. MDL-860 inhibited actinomycin D-resistant [3H]uridine uptake in cells infected with coxsackievirus A21 or rhinovirus 1-A, whereas incorporation into uninfected cell...

Density of infectious virus and complement-fixing antigens of two rhinovirus strains. J. Bacteriol. 91:1605-1611. 1966.-Two rhinovirus serotypes (echovirus 28 and HGP) and poliovirus type 1 were banded by isopycnic centrifugation in cesium chloride. The rhinovirus virions had a ...
متن کاملIn vitro and in vivo activities of WIN 54954, a new broad-spectrum antipicornavirus drug.
WIN 54954 (5-[5-[2,6-dichloro-4-(4,5-dihydro-2-oxazolyl)phenoxy]pentyl]-3- methylisoxazole) is a new member of the class of broad-spectrum antipicornavirus compounds known to bind in a hydrophobic pocket within virion capsid protein VP1. In plaque reduction assays, WIN 54954 reduced plaque formation of 50 of 52 rhinovirus serotypes (MICs ranged from 0.007 to 2.2 micrograms/ml). A concentration ...
متن کاملEnhancement of enterovirus infectivity in vitro by pretreating host cell monolayers with the cationic polymer polyethyleneimine.
Laboratory strains of enteroviruses, as well as viruses isolated from raw wastewater, were found to exhibit enhanced infectivity in vitro when BGM cell monolayers were pretreated with the cationic polymer polyethyleneimine (PEI). Viruses were assayed by the cytopathic effect technique and as PFU under methylcellulose and agar overlays with monolayers treated with 0 to 5.0 x 10(-3)% (wt/vol) PEI...
متن کاملMechanism of enhancement of virus plaques by cationic polymers.
It has been assumed that plaque enhancement by cationic polymers is due to their binding of sulfated polysaccharides in agar. However, viruses that are enhanced by cationic polymers, diethylaminoethyl-dextran, and protamine were found not to be inhibited by polyanions in agar under the usual overlay conditions. In the case of adenovirus, enhancement by protamine seems to be due to the protamine...
